Annotation Tato práce je zaměřena na způsob stanovení vitaminu D pomocí HPLC. V první části je popsán mechanismus vzniku tohoto lipofilního vitaminu a jeho fyziologie. Další část zpracovává princip metody HPLC. Třetí část je pak zaměřena na využití této metody při stanovení vitaminu D v konkrétních surovinách či potravinách. Význam vitaminu D spočívá v účasti při vstřebávání vápníku a fosfátu ze střeva a přispívá tak k regulaci a optimalizaci jejich hladiny v krvi. HPLC je jedna z nejpoužívanějších metod při stanovování všech vitaminů, tedy i vitaminu D. Jedná se o separační fyzikálně-chemickou metodu, která slouží k rozdělení složek vzorku rozpuštěného v roztoku mezi mobilní a stacionární fázi.
Annotation in English The aim of this thesis is focused on method of determination of vitamin D by HPLC. In the first part mechanism of origin this liposoluble vitamin and its physiology is described. Next part describes principle of HPLC method. The third part is focused on using this method in determination of vitamin D in raw materials or foods. The importance of vitamin D consists in attendance in absorption of calcium and phosphate from intestine and support regulation and optimalization of their level in blood. HPLC is one of the most widely used method to determination all of the vitamins. It is separative physical-chemical method, which is used to separate components in sample, which is dissolved in solution, between mobile and stationary phases.
