Práce se zabývá úpravou materiálových soustav na syntetické či přírodní bázi do podoby hierarchicky strukturovaných scaffoldů a tenkých vrstev. Pro tyto účely byly používány metody aditivní výroby, electrospinningu, fázové separace a leptání. Upravovanými materiály byly fotovytvrditelné pryskyřice, polykaprolakton, poly(vinylidenfluorid-trifluorethylen-chlortrifluorethylen), fibroin, polystyren a hliník. Vybrané substráty byly testovány a charakterizovány z hlediska materiálových i biologických vlastností. Výsledky testů buněčné kompatibility prokázaly zásadní vliv topografie povrchu na buněčnou proliferaci a v případě hierarchicky strukturovaných povrchů i na změnu morfologie kmenových buněk. Pro testování připravených strukturovaných povrchů s ohledem na buněčné chování v simulovaných in vivo podmínkách byl vyvinut nový typ průtokového bioreaktoru. Provedené experimenty poukázaly na zásadní vliv vznikajících nepravidelností v toku na buněčnou adhezi, na jehož základě byla provedena studie vlivu rozdílných typů toku na stabilitu polymerních povrchů.

This work is focused on modification of material systems on a synthetic or natural basis in the form of hierarchically structured scaffolds and thin layers. For this purpose, the methods of additive manufacturing, electrospinning, phase separation, etching were used. The materials treated were photocurable resins, polycaprolactone, poly (vinylidene fluoride-trifluoroethylene-chlorotrifluoroethylene), fibroin, polystyrene and aluminum. Not only the material properties, but also the cytocompatibility was evaluated. The results of showed a fundamental impact of surface topography on cell proliferation and, in the case of hierarchically structured surfaces, on the change in stem cell morphology. A new type of bioreactor was developed to test the prepared structured surfaces with respect to study of cell behavior under simulated in vivo conditions. The results confirmed the fundamental influence of flow irregularities on cell adhesion, on the basis of which a study of the influence of different types of flow on the stability of polymer surfaces was performed.
