Annotation Cílem této diplomové práce bylo studium dekarboxylázové aktivity kmenů izolovaných ze 3 druhů kefírů a jejich kefírových zrn (mléčné kefíry Manta a Flower a vodní kefír). Jednotlivé kmeny byly identifikovány metodou MALDI/TOF MS. Celkem bylo testováno 171 kmenů, z toho bylo zastoupeno 13 kmenů z čeledi Enterobacteriaceae, 62 kmenů kvasinek a 96 kmenů z rodu Lactobacillus. Byla zkoumána produkce 8 biogenních aminů (tryptaminu, 2-fenylethylaminu, putrescinu, kadaverinu, histaminu, tyraminu, spermidinu a sperminu). Pro stanovení koncentrací biogenních aminů byla využita metoda HPLC - UV/VIS. U všech testovaných kmenů byla zjištěna produkce 2-fenylethylaminu, putrescinu, kadaverinu, tyraminu a sperminu. Schopnost produkce histaminu byla prokázána pouze u 10 izolátů. U žádného kmene nebyl detekován tryptamin ani spermidin.
Annotation in English The aim of this thesis was to study the decarboxylase activity of strains isolated from 3 kinds of kefir and their kefir grains (milk kefir Manta and Flower and water kefir). Individual strains were identified by the MALDI/TOF MS method. A total of 171 strains were tested, of which 13 strains of Enterobacteriaceae, 62 strains of yeast and 96 strains of Lactobacillus were represented. The production of 8 biogenic amines (tryptamine, 2-phenylethylamine, putrescine, cadaverine, histamine, tyramine, spermidine and spermine) was investigated. The HPLC - UV/VIS method was used to determine the concentrations of biogenic amines. All strains were found to produce 2-phenylethylamine, putrescine, cadaverine, tyramine and spermine. Only 10 isolates have been shown to produce histamine. No strain produced tyramine or spermidine.
