Obsahem teoretické části diplomové práce je zopakování základů tuků a olejů. Značná část se skládá z popisu plynové chromatografie, která byla při praktické části využívána jako analytická koncovka. Pro následné využití plynové chromatografie byla popsána kapitola derivatizace vzorku. V poslední kapitole byly popsány jednotlivé metody derivatizace (kyselá a bazická), ke kterým byly přiřazeny mechanismy esterifikačních a transesterifikačních reakcí.
V praktické části byly popsány bazicky katalyzované esterifikace s hydroxidem draselným a methanolátem sodným. Za skupinu kysele katalyzované esterifikace byly popsány esterifikace s kyselinou sírovou a fluoridem boritým. Jako modelové vzorky byly použity komerčně dostupné oleje olivový, řepkový a slunečnicový. Všechny vzorky byly po derivatizaci analyzovány pomocí plynové chromatografie a následně zhodnoceny a mezi sebou porovnány.

The content of the theoretical part of diploma thesis is repetition of basic information of fats and oils. A large part consists of the description of gas chromatography, which was used in the practical part like a analytical tip. For subsequent use of gas chromatogramy has been described chapter derivatization samples. In the last chapter were described the individual methods of derivatization (acidic and basic) to which the mechanisms of esterification and transesterification reactions have been assigned.
In the practical part was described basic catalyzed esterification with acid, hydroxide and sodium methoxide. In the group of the acid catalyzed esterification was described esterification with sulfuric acid and boron fluoride. As the model samples were used comercially oils olive, rapeseed and sunflower oils. All samples were analyzed by gas chromatography after derivatization and subsequently evaluated and compared.
