Annotation Diplomová práce se zabývá problematikou kumarinových sloučenin, jejichž vlastnosti a syntéza jsou popsány v teoretické části. Zmíněny jsou také některé biologické aktivity. V praktické části jsou popsány pokusy o přípravu 3-substituovaných 4-hydroxykumarinů jakožto analogů k derivátům 4-hydroxychinolin-2-onům. Experimenty byly založeny na vysokoteplotní kondenzaci fenolu s diethylestery kyseliny malonové. S připraveným 3 fenyl-4-hydroxykumarinem byly dále provedeny pokusy s halogenačními činidly, jež měly směřovat k 3-halogenchroman-2,4-dionům. Neočekávaně ale byly získány deriváty ethanonu. Blíže bylo prozkoumáno chování 2-chlor-2-fenyl-1-(2-hydroxyfenyl)ethanonu v bazickém prostředí.
Annotation in English This diploma thesis is focused on coumarin compounds, their properties and synthesis are covered in the theoretical part. Some biological activities are also mentioned. In the practical part, attempts to prepare 3-substituted 4-hydroxycoumarins as analogs to 4-hydroxyquinolin-2-one derivatives have been described. The experiments were based on the high-temperature condensation of phenol with diethyl esters of malonic acid. With the prepared 3-phenyl-4-hydroxycoumarin, experiments targeting to 3-halogenchroman-2,4-diones were further performed. Unexpectedly, ethanone derivatives were obtained. The reactivity of 2-chloro-1-(2-hydroxyphenyl)-2-phenyl-ethanone in a basic environment was further investigated.

Research Plan I. Teoretická část
  1. Zpracovat literární rešerši zaměřenou na přípravu a biologické účinky analogů k připraveným derivátům v rámci DP.
II. Praktická část
  1. Pokusit se nalézt vhodnou metodu pro přípravu sloučenin s 4-hydroxykumarinovým skeletem.
  2. Provést pokusy o přeměnu 4-hydroxykumarinů na 3-halogen- případně na 3-aminoderiváty chroman-2,4-dionu.
  3. Izolované produkty charakterizovat dostupnými metodami instrumentální analýzy.
  4. Vyhodnotit výsledky experimentů a sepsat diplomovou práci.
Research Plan
I. Teoretická část
  1. Zpracovat literární rešerši zaměřenou na přípravu a biologické účinky analogů k připraveným derivátům v rámci DP.
II. Praktická část
  1. Pokusit se nalézt vhodnou metodu pro přípravu sloučenin s 4-hydroxykumarinovým skeletem.
  2. Provést pokusy o přeměnu 4-hydroxykumarinů na 3-halogen- případně na 3-aminoderiváty chroman-2,4-dionu.
  3. Izolované produkty charakterizovat dostupnými metodami instrumentální analýzy.
  4. Vyhodnotit výsledky experimentů a sepsat diplomovou práci.
